What supplements would you most like to see be made for Guardians?
I've been considering some class specific, like a Book of Bruisers with several example NPC's and some expanded rules for your Bruiser character. The superhero genre is so vast there is no limit to what you can do!
Would a settings book be of interest?

Love to see an "Advanced" Guardians book of optional rules, erratta, new powers, new classes, etc... Nothing that invalidates the old book, but kind of like an "Expert Set" add on to the basic rules.

What additional classes could be added that aren't covered in the core book?
I can see a need for a sorcerer class to cover characters like Dr. Strange, Zatanna, Raven, Scarlet Witch, Magik, Wiccan, Etrigan The Demon, Brother Voodoo, Dr. Fate, or The Spectre.
At the time Guardians was written it was seen as just another component within the general OSR ecosphere. So you could port in a traditional magic-user from any other Basic-compatible RPG. Adding in magic and a bunch of spells would have blown up the size of the book. So it kind of deserves its own supplement.

I love all of these ideas, and aa71 above is really on point with his suggestion as well.... please, yes! Another thing, since stats can run so high and this almost make Saves or Skill tests moot, maybe a DC system to switch out to d100's for testing instead of d20?

Yes, not a ton of classes... You can do quite a bit with the rules as currently presented. Maybe a sorcerer, and perhaps the Vigilante class that was posted on a blog somewhere. Maybe 1 or 2 others might come to light, but can't think of any right now.

Also love to see some new powers, new limits, new issues, more vehicles, weapons, equipment, etc new vehicles, weapons, and equipment sound like good ideas. The powers, limits, and issues were carefully designed around each other in the rule book and are much harder to add to without disrupting the balance (such as it is in a superhero game). Not saying it can't or won't be done, just that the design space is much more restricting.

Setting books would be nice. Especially a WW2 supers book.
